腺嘌呤致慢性肾功能衰竭大鼠模型制备及钙磷代谢变化

Establishment of rats model of chronic renal failure induced by adenine and changes of calcium and phosphorus metabolism

摘要: 目的 研究慢性肾功能衰竭(CRF)大鼠模型的制备及其钙磷代谢变化。方法 将80只SD雄性大鼠随机分为正常对照组(22只)和CRF组(58只),正常对照组大鼠用普通饲料喂养,CRF组用含0.75%腺嘌呤饲料喂养,连续6周。观察两组大鼠一般状态的改变,检测其血肌酐(SCr)、尿素氮(BUN)、血钙和血磷、24h尿钙和尿磷、24h粪钙及校正体重后的24h尿钙和尿磷、24h粪钙水平,以肉眼和显微镜观察两组大鼠肾脏组织的大体和病理变化。结果 CRF组大鼠血BUN和SCr水平均高于正常对照组[(55.90±11.54)mmol/L比(7.18±2.12)mmol/L,(232.47±60.83)μmol/L比(68.47±16.24)μmol/L],体重及血钙水平均低于正常对照组[(180.59±23.58)kg比(400.63±18.46)kg,(2.26±0.32)mmol/L比(2.54±0.31)mmol/L,P<0.05]。CRF组大鼠24h尿钙及尿磷代谢水平明显高于正常对照组[(0.76±0.30)mg/d比(0.41±0.23)mg/d,(0.70±0.31)mg/d比(0.25±0.21)mg/d],校正体重后的24h尿钙及尿磷、粪钙代谢水平也均高于正常对照组 [(4.49±2.12)mg·kg-1·d-1比(1.01±0.55)mg·kg-1·d-1, (556.08±225.03)mg·kg-1·d-1比(237.92±104.77)mg·kg-1·d-1,(3.92±1.48)mg·kg-1·d-1比(0.52±0.41)mg·kg-1·d-1,P<0.01]。CRF组大鼠的肾脏病理组织变化以肾小管间质损害为主。结论 0.75%腺嘌呤饲料混合喂养大鼠可以诱导CRF模型,并出现CRF的生化及肾小管间质病理改变,从而影响钙磷代谢。

Abstract: Objective To study the establishment of rats model of chronic renal failure(CRF) and observe the changes of calcium(Ca) and phosphorus(P) metabolism.Methods SD male rats(80 cases) were randomly divided into normal control group(22 cases) and CRF model group(58 cases).The normal control group were fed a normal diet,while the CRF group were fed with 0.75% adenine diet for 6 weeks.The changes of the general state of the rats were observed.The levels of serum creatinine(SCr),blood urea nitrogen(BUN),serum Ca and P,24h urine Ca and P,24h fecal Ca and the levels of 24h urine Ca and P,24h fecal Ca after weight correction were detected.The gross and pathological changes of the renal tissues of the 2 groups were observed by naked eye and microscope.Results The levels of BUN and SCr in CRF group were significantly higher than those in normal control group[(55.90±11.54)mmol/L vs (7.18±2.12)mmol/L,(232.47±60.83)μmol/L vs (68.47±16.24)μmol/L],the weight and blood Ca levels were lower than those in the normal control group[(180.59±23.58)kg vs (400.63±18.46)kg,(2.26±0.32)mmol/L vs (2.54±0.31)mmol/L,P<0.05].The metabolism of 24h urine Ca and P in CRF group were higher than those in the normal control group[(0.76±0.30)mg/d vs (0.41±0.23)mg/d,(0.70±0.31)mg/d vs (0.25±0.21)mg/d],the metabolism of 24h urine Ca and P,fecal Ca in CRF group were higher than those in normal control group [(4.49±2.12)mg·kg-1·d-1 vs (1.01±0.55)mg·kg-1·d-1,(556.08±225.03)mg·kg-1·d-1 vs (237.92±104.77)mg·kg-1·d-1,(3.92±1.48)mg·kg-1·d-1 vs (0.52±0.41)mg·kg-1·d-1P<0.01].The pathological changes of kidney in CRF group were mainly tubulointerstitial lesion.Conclusion The CRF model is successfully induced by 0.75% adenine in rats.There are CRF biochemical and pathological changes of renal interstitial tubules,which affects the metabolism of Ca and P.
